Abstract:
To clarify the application of the lipase from
Moesziomyces aphidis, this research was conducted to investigate the effects of reaction temperature, pH and stability, the effects of metal ions, surfactants, inhibitors and organic solvents on the activity of lipase. The results showed that the optimum reaction temperature and pH value of the lipase were 70 ℃ and 8, respectively, this lipase had good temperature stability and pH tolerance, and it was a kind of high temperature resistant alkaline lipase synthesized by room temperature fermentation. Zn
2+ had a significant inhibitory effect on its activity. Surfactants such as Tween-20, Tween-80, Triton 100 and organic solvents such as methanol, ethanol, n-propanol, isopropanol and acetone showed significant promotion effect on lipase activity. The lipase was able to hydrolyze both the 1,3 and 1,2 ester bonds of triglycerides, the kinetic parameters
Km and
vmax for hydrolysis of
p-NPP were 1.14 mmol/L and 119.05 mol/min, respectively. Therefore, the synthetic lipase of
Moesziomyces aphidis has good prospects for application under industrial reaction conditions such as high temperature, weak alkalinity and organic solvents.
