麦胚无细胞蛋白合成系统研究进展

    Research progress on the wheat germ cell-free protein synthesis system

    • 摘要: 无细胞蛋白合成(CFPS)系统作为基础研究、应用科学和产品开发的通用工具受到广泛关注,利用CFPS系统合成目标蛋白在食品、医药以及合成生物学领域取得了巨大进展。在现有的CFPS系统中,麦胚无细胞蛋白合成(WG-CFPS)系统因具有原料来源广泛、蛋白产率高、适合表达复杂真核蛋白及膜蛋白等特点,已被作为高性价比的潜力对象用于商业化开发。综述了与WG-CFPS系统相关的研究现状,总结并讨论了该系统下的各种优化策略及优化结果。针对当前WG-CFPS系统在食品、医药等领域的应用进行了归纳和梳理,并对其未来的挑战和发展进行了展望。

       

      Abstract: The cell-free protein synthesis (CFPS) system has received widespread attention as a general tool for basic research, applied science, and product development. Significant advancements have been achieved in synthesizing target proteins using CFPS systems in the fields of food, medicine, and synthetic biology. Among the existing CFPS systems, the wheat germ-based cell-free protein synthesis (WG-CFPS) system stands out due to its wide source of raw materials, high protein yield, and suitability for expressing complex eukaryotic proteins and membrane proteins. It has been identified as a cost-effective and promising candidate for commercial development. This review summarizes the current research status related to the WG-CFPS system and discusses various optimization strategies and their outcomes within this system. In addition, the current application of WG-CFPS system in food, medicine and other fields is summarized and sorted out, and its future challenges and development are prospected.
