高聚合度麦麸阿魏酸糖酯的组成特征及其自由基清除能力研究

    COMPOSITION CHARACTERISTICS AND RADICAL SCAVENGING ACTIVITY OF WHEAT BRAN FERULOYLATED GLYCOSIDES WITH HIGH POLYMERIZATION DEGREE

    • 摘要: 分离纯化麦麸阿魏酸糖酯,对其高聚合度组分的组成特征进行分析,并研究其自由基清除能力。采用酸水解法制备麦麸阿魏酸糖酯,经纯化后以Sephadex LH-20凝胶色谱柱分离得到高聚合度组分F1,用乙酰丙酮法测定其平均聚合度,用高效液相色谱法(HPLC)检测组分中的阿魏酸,用气相色谱法(GC)检测糖体组成,用红外光谱法(IR)检测其糖苷键的组成与类别并分析结构特征,用分光光度法检测其对DPPH和·OH自由基的清除能力。结果表明:经纯化、分离得到不同聚合度的麦麸阿魏酸糖酯,对其高聚合度组分F1的结构分析可知,其含有酯化的阿魏酸、阿拉伯糖和木糖结构,具有β(1-4)吡喃糖糖苷键,其平均聚合度为10.9;F1对DPPH和·OH自由基的清除能力与其质量浓度呈正相关,其中各浓度下对·OH自由基的清除效果均优于游离阿魏酸。高聚合度麦麸阿魏酸糖酯组分F1为阿魏酰基阿拉伯低聚木糖,由阿魏酸与阿拉伯木聚糖酯化构成,具有良好的DPPH和·OH自由基清除能力,极具研究前景。

       

      Abstract: The present study was to investigate the composition characteristics and free radical scavenging activity of wheat bran feruloylatedgl ycosides with high polymerization degree.Feruloylated glycosides of wheat bran was prepared by acid hydrolysis method,and then the component of F1 with high polymerization degree was purified and isolated by Amberlite XAD-2 macroporous resin and Sephadex LH-20 gel chromatography.The average polymerization degree of F1 was determined by the acetylacetone method;phenolic acid (ferulaic acid) were detected by high performance liquid chromatography (HPLC);sugar composition was examined by gas chromatography (GC);and the glycosidic linkage was measured by infrared spectroscopy (IR).Moreover,the scavenging capacity of F1 on DPPH and·OH was detected by using the UV spectrophotometry method and compared with ferulic acid standard.The feruloylated glycosides of wheat bran with different polymerization degree were obtained by separation and purification.The composition analysis of F1 with high polymerization degree showed that esterified ferulic acid,arabinose and xylose with β(1-4) pyranoside bond were contained,and the average polymerization degree was 10.9.Furthermore,the scavenging capacity of F1 on·OH and DPPH had positive correlation with the concentration,in which the·OH scavenging ability of F1 was better than that of free ferulic acid.Briefly,the component of F1 with high polymerization degree was feruloylated arabinoxylan,which was composed of ferulic acid and arabinoxylan.F1 has better free radical scavenging capacity on DPPH and·OH,which will have a wide research and development prospect.
