超声波辅助双水相提取胡萝卜叶总黄酮的工艺研究

    OPTIMIZATION OF ULTRASONIC-ASSISTED AQUEOUS TWO-PHASE EXTRACTION OF TOTAL FLAVONOIDS FROM CARROT LEAVES

    • 摘要: 胡萝卜叶有香气而不易感染病虫害,是非常好的绿色食品原料。为提高胡萝卜叶的开发利用率,以胡萝卜叶为原料,优化提取胡萝卜叶中总黄酮的工艺条件。以总黄酮提取率为指标,采用超声波辅助双水相对胡萝卜叶中的总黄酮进行提取,在单因素的基础上,采用Box-Behnken响应面法对影响胡萝卜叶总黄酮的提取工艺条件进行优化,建立了乙醇体积分数、超声温度、液料比、硫酸铵质量浓度四因子与总黄酮提取率的二次多项式回归方程。单因素试验结果表明,总黄酮的提取率随着乙醇体积分数、超声温度、液料比和硫酸铵质量浓度的增加呈现先增加后减小的变化趋势。响应面优化超声辅助双水相提取胡萝卜叶总黄酮的最佳提取工艺条件为:乙醇体积分数 64.39%,提取温度61.32℃,液料比34.48 mL/g和硫酸铵质量浓度0.41 g/mL。经修正后的最佳工艺条件为:乙醇体积分数 64%,超声温度61℃,液料比34 mL/g和硫酸铵质量浓度0.41 g/mL,在此条件下,测得胡萝卜叶总黄酮的提取率为4.956%,与理论值相比,其相对误差为0.52%,验证了回归模型的有效性。利用Box-Behnken响应面法优化超声波辅助双水相提取胡萝卜叶总黄酮的工艺准确率高,可靠性强。

       

      Abstract: Carrot leaves have aroma and are not easy to be infected by pests and diseases,which are one kind of good raw material for green food.In order to improve the utilization of carrot leaves,the extraction process conditions of total flavonoids from carrot leaves were optimized by ultrasonic-assisted aqueous two-phase extraction technology with the extraction yield as evaluation index.Based on the single factors experiments,the involving factors were optimized using Box-Behnken response surface methodology and a quadratic polynomial regression equation was established for the extraction yield of total flavonoids and ethanol concentration,ultrasonic temperature,liquid to material ratio and ammonium sulfate concentration.Experimental results of single factors showed that the extraction yield of total flavonoids increased firstly and dropped subsequently with the increase of the above four related factors.The optimum extraction conditions of total flavonoids from carrot leaves by the ultrasonic-assisted aqueous two-phase method were as follows:ethanol concentration of 64.39%,ultrasonic temperature of 61.32℃,liquid to material ratio of 34.48 mL/g and ammonium sulfate concentration of 0.41 g/mL.The revised optimum process conditions were:ethanol concentration of 64%,ultrasonic temperature of 61℃,liquid to material ratio of 34 mL/g and ammonium sulfate concentration of 0.41 g/mL.The extraction yield of total flavonoids could be up to 4.956% and the relative error was 0.52% compared to the predictive value,which testified that the regression model was fitted well with the experimental data.It was concluded that ultrasonic-assisted aqueous two-phase extraction of total flavonoids from carrot leaves optimized by Box-Behnken response surfacemethodology had high accuracy and reliability.
