Abstract:
The oligosaccharide extracted from wheat bran have the functions of promoting probiotic proliferation,enhancing immune response,inhibiting oxidative stress induced by high fat diet,moisturizing intestines and relieving defecation,and decreasing blood glucose.In order to improve the production efficiency of wheat bran oligosaccharides,the effect of pretreatment conditions of wheat bran,xylanase addition amount,enzymatic hydrolysis pH,enzymatic hydrolysis time and enzymatic hydrolysis temperature on the xylanolysis amount of wheat bran were studied by single factor experiment at different temperatures above 150℃.The effect of activated carbon amount,decolorization time and decolorization temperature on the decolorization rate of sugar solution were also studied.The results showed that the optimum cooking conditions above 150℃ were as follows:acetic acid content 0.15% of dry weight of wheat bran,cooking temperature 160℃ and cooking time 20 min.Under these conditions,the yield of soluble pentosan was the highest,reaching 15.5% of dry weight of wheat bran,the color was light and the harmful substance furfural content was also less.The optimum technological parameters of xylanase hydrolysis were as follows:xylanase addition 0.20%,enzymatic hydrolysis pH 5.0,enzymatic hydrolysis time 4 h and enzymatic hydrolysis temperature 45℃,under which the dissolution rate of soluble pentosan was up to 27.70 g/L.The optimum decolorization parameters were as follows:the amount of activated carbon 6%,the decolorization temperature 60℃ and the decolorization time 50 min,under which the decolorization rate of sugar solution reached to 97% and the color of afforded decolorized sugar solution was clarified.The optimized technological conditions have high production efficiency but low cost,which can be used as reference for the wheat bran oligosaccharides production.
