超声波-纤维素酶法提取板栗壳中原花青素及其提取液抗氧化活性分析

    Analysis of Ultrasonic-Enzymatic Extraction of Procyanidins from Chestnut Shell and Antioxidant Activities of Their Extracts

    • 摘要: 为分析超声波-纤维素酶法联合提取对板栗壳中原花青素的提取量及提取液抗氧化活性的影响,在单因素试验的基础上通过正交试验,对板栗壳原花青素的超声波法和纤维素酶法提取条件进行优化,对比超声波法、酶法、先超声波后酶法、先酶法后超声波法以及超声波酶法同步这5种方法对板栗壳中原花青素的提取量的影响,并分析其提取液抗氧化能力。结果表明:超声波法在最佳提取条件下得到的原花青素提取量为21.03 mg/g;酶法在最佳提取条件下得到的原花青素提取量为10.12 mg/g;采用先超声波后纤维素酶法及先纤维素酶法后超声波法得到的原花青素提取量分别为23.58、19.04 mg/g;对比得出超声波纤维素酶法同步提取的原花青素的提取量最高,为26.46 mg/g,且通过此方法得到的提取液具有良好的抗氧化活性,DPPH自由基的清除率为74.11%,羟基自由基清除率为55.21%。试验证明超声波纤维素酶法同步进行提取得到的板栗壳原花青素的提取量最高,提取液抗氧化活性最好,可为板栗壳的回收利用提供理论依据。

       

      Abstract: The effect of combined ultrasonic-cellulase extraction on the extraction amount of procyanidins (PCs) from chestnut shell and the antioxidant activity of the extract was analyzed. The optimization of PCs extraction from chestnut shell by ultrasound and cellulase extraction condition were investigated by single factor and orthogonal experiments. Furthermore, the effects of ultrasonic-assisted extraction, cellulase-assisted extraction, and their sequential and simultaneous combinations on the extraction efficiencies of PCs and the antioxidant activities of the resulting extracts were evaluated. The results showed that the extraction amount of PCs obtained by ultrasonic and cellulase extraction methods under the optimal extraction conditions were 21.03 mg/g and 10.12 mg/g, respectively. The extraction amount of PCs obtained by ultrasonic-cellulase extraction and cellulase-ultrasonic extration were 23.58 and 19.04 mg/g, respectively. The simultaneous ultrasonic and enzymatic treatment could obtain the highest conten of PCs, which was 26.46 mg/g. Furthermore, the DPPH radical scavenging ability and hydroxyl radical scavenging ability of the obtained extracts was also the highest, with values of 74.11% and 55.21%, respectively. This research results confirmed that the extraction amount of PCs obtained by ultrasonic and cellulase methods was the highest and the antioxidant activities of the extract was the best. It provided reliable theory basis for the recycling of chestnut shell.
