响应面法优化玉米叶酸提取工艺的研究

    Optimization of Extraction Process of Folic Acid from Maize by Response Surface Methodology

    • 摘要: 玉米是我国主粮作物之一,富含各种维生素等营养物质。通过单因素试验考察提取时间、处理温度、料液比和活性炭对叶酸提取过程的影响,并用高效液相色谱进行检测。利用Box-Behnken试验设计优化叶酸提取的最佳工艺条件为提取温度42℃、料液比1:11.7 g/mL、提取时间7.3 h、活性炭用量1.2 g,在此条件下进行3次平行试验,测得的玉米中叶酸含量为(1.68±0.05)μg/g。分析表明4个因素中温度对叶酸的提取影响最大。该研究以期为主粮作物中叶酸含量的研究提供参考。

       

      Abstract: Maize is one of the main grain crops in China, which is rich in various vitamins and other nutrients.The influences of extraction time, treatment temperature, solid-to-solvent ratio and activated carbon load on the extraction of folic acid were investigated by the single factor experiments and detected by high performance liquid chromatography.By Box-Behnken methodology, the extraction conditions were optimized as follows:extraction temperature 42℃, solid-to-solvent ratio 1:11.7 (g/mL), extraction time 7.3 h and activated carbon load 1.2 g.Three parallel experiments were performed under the optimized conditions, and the content of folic acid in maize was 1.68±0.05 μg/g.Results also indicated that, among the four factors, the effect of temperature was the most significant.These can provide the available information for the study of folic acid content in main grain crops.
