细菌纤维素生产菌的常压室温等离子体诱变育种及发酵条件优化

    BREEDING OF BACTERIAL CELLULOSE-PRODUCING BACTERIA UNDER ARTP MUTATION AND OPTIMIZATION OF FERMENTATION CONDITIONS

    • 摘要: 采用常压室温等离子体(ARTP)诱变技术对目的菌株Komagataeibacter xylinus进行诱变,选育BC高产株,经10次传代发酵验证其遗传性较稳定。在单因素试验基础上,以BC产量为响应值,采用Plackett-Burman、Central composite design试验设计法进行响应面分析。结果表明,高产菌株G31的最佳发酵工艺条件为葡萄糖∶甘露醇(1∶1)4.0 g·(100 mL)-1、MgSO4·7H2O 0.13 g·(100 mL)-1、冰乙酸 0.25 mL·(100 mL)-1。在此发酵条件下,BC产值为 8.49 g·L-1,与预测值 8.61 g·L-1相差不大,是初始菌株发酵产量的2.53倍。

       

      Abstract: Mutagenesis of the target strain Komagataeibacter xylinus was carried out using ARTP mutagenesis technique.The BC high-yield strain,named G31,was selected and its heredity was stable after 10 subcultures.On the basis of single factor experiments,response surface analysis was performed using Plackett-Burman and Central composite design,while BC yield was as the response value.The results showed that the optimum fermentation conditions for the high-yield strain G31 were as follows:the ratio of glucose to mannitol of 1:14 g·(100 mL)-1,MgSO4·7H2O 0.13 g·(100 mL)-1,and glacial acetic acid 0.25 mL·(100 mL)-1.Under the optimal fermentation conditions,the BC production value was 8.49 g/L,which had no obvious difference with the predicted value (8.61 g/L).In addition,the BC production of Mutagenesis strain was 2.63 times as much as the initial strain.
