赤拟谷盗触角电位测量方法优化及对小麦粉挥发物的触角电位反应

    Optimization of Electroantennogram Measurement Method and Electroantennogram Responses of Tribolium castaneum(Herbst) to Wheat Flour Volatiles

    • 摘要: 优化了赤拟谷盗触角电位(EAG)测量方法,用此法测试了赤拟谷盗对14种小麦粉挥发物的触角电位反应。结果表明,优化后的方法能获得稳定、可靠的EAG数据;赤拟谷盗触角对14种小麦粉挥发物的刺激均可产生反应,雌、雄虫对这些挥发物的触角电位反应趋势相似。赤拟谷盗对醛类小麦粉挥发物的触角电位反应最高,对烃类小麦粉挥发物的触角电位反应均处于较低的水平。引发赤拟谷盗雌雄虫最高触角电位值的前4种化合物分别是戊醛、壬醛、2-辛烯醛、2-戊基呋喃,雌雄虫对十四烷和2-辛烯醛的触角电位反应差异显著。解决了赤拟谷盗触角电位测试过程中触角与电极连接难的问题,为揭示赤拟谷盗的嗅觉反应特点提供了基础数据。

       

      Abstract: In order to obtain more reliable electroantennogram (EAG) of Tribolium castaneum (Herbst), the EAG measurement method was optimized and the EAG response of T. castaneum to 14 wheat flour volatiles was tested. The results showed that the optimized method obtained stable and reliable EAG data. All the 14 wheat flour volatiles stimulated EAG responses of T. castaneum, and the EAG responses of males and females to these compounds were similar. The EAG response to aldehyde from wheat flour volatiles was the highest, while the EAG response to hydrocarbon wheat flour volatiles was at a low level. The highest EAG of male and female was initiated by the same four compounds: valeraldehyde, nonanal, 2-octenaldehyde, and 2-pentylfuran. The EAG response values of males and females to tetradecane and 2-octenaldehyde were significantly different. The results solve the difficulty in connecting antennae to electrode during testing the EAG response of T. castaneum, and provide basic data to reveal the characteristics of the EAG response of T. castaneum.
