怀山药多糖的提取、硫酸酯化修饰及抗氧化活性研究

    Study on the Extraction,Sulfation Modification and Antioxidant Activity of Yam Polysaccharides

    • 摘要: 为了提高怀山药多糖的应用价值,从怀山药中提取并分离纯化山药多糖,采用气相色谱/质谱(GC/MS)、傅立叶红外光谱仪、多角度激光光散射仪(MALLS)和核磁共振技术对山药多糖结构进行初步表征,然后采用氯磺酸-吡啶法对山药多糖进行硫酸酯化修饰,利用傅立叶红外光谱仪鉴定修饰结果,最后测定多糖及其衍生物的抗氧化活性。结果表明:山药多糖中主要单糖成分为葡萄糖84.5%、木糖11.4%、半乳糖2.3%、阿拉伯糖1.4%;主要结构为C3和C4位置支化的β-葡聚糖,即β-1,3-葡聚糖、β-1,4-葡聚糖,另外含少量α-葡聚糖。山药多糖的分子质量为6.6×104 Da,而硫酸酯化山药多糖分子质量小于1.0×104 Da。在相同质量浓度下,硫酸酯化山药多糖的抗氧化活性高于山药多糖。

       

      Abstract: In order to improve the application value of polysaccharides from Chinese yam,the polysaccharides were extracted and isolated from the Chinese yam.Gas chromatography/mass spectrometry (GC/MS),fourier transform infrared spectroscopy,multi-angle laser light scattering (MALLS) and nuclear magnetic resonance (NMR) were used to characterize the structure of yam polysaccharides.Then,yam polysaccharides were sulfated by chlorosulfonic acid-pyridine method,and identified by FT-IR.Finally,the antioxidant activities of the polysaccharides and its derivatives were determined.The results showed that,the main monosaccharides in yam polysaccharides were 84.5% glucose,11.4% xylose,2.3% galactose and 1.4% arabinose.And the main structures were β-1,3-glucan and β-1,4-glucan.The content of α-glucan was low.The molecular weight of the yam polysaccharide was 6.6×104 Da.However,the molecular weight of sulfated yam polysaccharide was lower than 1.0×104.At the same mass concentrations,the antioxidant activities of sulfated yam polysaccharides were higher than that of yam polysaccharides.
