    基于胶体金-适配体的层析试纸条检测玉米中黄曲霉毒素B1

    Detection of aflatoxin B1 in corn by chromatography strip based on colloidal gold aptamer

    • 摘要: 黄曲霉毒素B1 (AFB1)因其剧毒性及在粮食中分布广泛而成为人类健康和粮食安全的重大风险因素之一,因此建立快速、灵敏的检测方法十分必要。利用胶体金和适配体作为金标探针,构建一种新型层析试纸条以实现玉米中AFB1的快速检测。根据体系中AFB1存在会减弱生物素标记的适配体与金标探针的结合,从而导致试纸条检测线的颜色强度降低或消失,进而实现对AFB1高效灵敏检测。在最佳条件下,试纸条检测AFB1的检出限值低至0.5 ng/mL,在0.5~500 ng/mL范围内快速准确检测且对AFB1具有高度的特异性。本研究制备的适配体试纸条能够成为玉米中AFB1检测的有效方法,具有良好的应用前景。

       

      Abstract: Aflatoxin B1 (AFB1) has become one of the significant risk factors for human health and food security due to its hypertoxicity and wide distribution in grain. Therefore, it is necessary to detect it. In this study, a novel chromatographic test strip (AuNPs-LFAA) was constructed for the rapid detection of AFB1 in maize using colloidal gold (AuNPs) and aptamer as the nano-golden probe. Based on the fact that the presence of AFB1 in the system attenuates the binding of biotin-modified aptamers (Bio-Apt) and thiol-modified complementary chain covalently bound AuNPs (AuNPs-SHcDNA) will be weakened, thusthe color intensity of the test line of the test strip would be reduced.Therefore, how to achieve efficient and sensitive detection of AFB1 is required. Under the optimal conditions, AuNPs-LFAA detected AFB1 with a limit of detection (LOD) as low as 0.5 ng/mL, providing a rapid and accurate detection in the range of 0.5-500 ng/mL with high specificity and sensitivity for AFB1. In summary, AuNPs-LFAA prepared in this study can be an effective method for the detection of AFB1 in maize and has good application prospects.
